Production of an E. coli Toxin Protein; Colicin A in E. coli Using an Inducible System

چکیده
The ultimate aim of biochemistry is to decipher how events occur inside of living cells or organisms. The living cell must also be regarded as the ideal model system for any biochemical application, as most physiological targets are found inside living cells. The cell envelope of the Gram-negative bacterium Escherichia coli is a complex structure and many of the proteins found in the bacterial envelope are involved in import mechanisms. A variety of bacterial toxins have taken advantage of some of these envelope proteins to enter the bacteria. Pore-forming toxins present the important challenge of the translocation process, and the study of the membrane insertion mechanism of pore-forming colicins has also become a very important subject. How these proteins pass through or integrate into membranes is still one of the fundamental unsolved problems in biochemistry. Colicins are plasmid-encoded toxin proteins produced by immune E. coli that are active against sensitive E. coli and closely related cells. Their toxic activities are of various types; some colicins form ion ∗Corresponding author

Extraction, Cloning and Expression of RTB, as a Vaccine Adjuvant/Carrier,in E. coli and Production of Mouse Polyclonal Antibody (Anti-B chain Abs)
Ricin, the toxic lectin extracted from the castor bean plant (Ricinus communis), consists of an A chain (RTA) and a B chain (RTB). Anti-A chain Abs and anti-B chain Abs can neutralize toxins in vivo and in vitro via blocking the binding of the toxin to the cell. Also, RTB protein is able to serve as an antigen deliver to the mucosal immune system and act as an immunoad...
متن کاملExpression, purification, and immunization of a chimeric protein containing immunogenic regions of flagellin and intimin proteins against E. coli O157: H7
Introduction: Enterohemorrhagic Escherichia coli (EHEC) and serotype O157: H7 is one of the most important diseases causing diarrhea. Shiga-like toxin secreted by the bacteria destroys epithelial cells and, in acute cases, causes hemolytic uremic syndrome (HUS). Antibiotic therapy is not effective against this pathogen, because it increases the production of Shiga toxin. Designing chimeric immu...
متن کاملProduction of Recombinant Proline Dehydrogenase Enzyme from Pseudomonas fluorescens pf-5 in E. coli System
Proline dehydrogenase (ProDH; 1.5.99.8) belongs to superfamily of amino acid dehydrogenase, which plays a significant role in the metabolic pathway from proline to glutamate. The goal of this research was gene cloning and characterization of ProDH enzyme from Pseudomonas fluorescens pf-5 strain. The gene encoding ProDH was isolated by means of PCR amplification and cloned in an IPTG inducible T...
متن کاملRecombinant Production of a Novel Fusion Protein: Listeriolysin O Fragment Fused to S1 Subunit Of Pertussis Toxin
Background: Some resources have suggested that genetically inactivated pertussis toxoid (PTs) bear a more protective effect than chemically inactivated products. This study aimed to produce new version of PT, by cloning an inactive pertussis toxin S1 subunit (PTS1) in a fusion form with N-terminal half of the listeriolysin O (LLO) pore-forming toxin. Methods: Deposited pdb structure file of the...

Background and purpose: The most common cause of diarrhea is Shigella and no vaccine has been found so far. IpaD and STxB proteins (B subunit of Shiga toxin) play an important role in invasion, infection and pathogenesis caused by Shigella. To evaluate the immunogenicity of each of the proteins IpaD and STxB can using of two animal models mice and guinea pigs and could be determined role of eac...
